The aim of this study was to investigate the effect of bovine serum albumin (BSA), lecithin and trehalose on the qualitative indices of freeze-thawed Acipenser persicus sperm. Sperm samples of five males were diluted (1:3) with an extender containing (0.2 mM glucose, 10% methanol) with BSA, soybean lecithin and trehalose diluted to 1% final volume by manual freezing method. After 3 months of freezing, sperm quality indices were evaluated by Image J software and mitochondrial activity by JC_1 method. Results of sperm motility showed rotational motions in treatment 2 and linear motions in treatment 4. Analysis of motion parameters showed that the treatment 4 had the highest arc velocity (1.74 μM/s) and highest linear velocity were in treatme
The aim of this study was to investigate the effect of bovine serum albumin (BSA), lecithin and trehalose on qualitative indices of freeze-thawed Acipenser persicus sperm. In this regard, sperm samples prepared from 5 male A. persicus and the samples were frozen after dilution with various extenders. After 3 months of freezing, the freeze-thawed sperm quality indices were evaluated by Image J software and mitochondrial activity by using the fluorescent staining JC-1 method. Results of sperm motility showed rotational motions in treatment BSA and linear motions in treatment “BSA + trehalose”. Analysis of motion parameters in freeze-thawed sperm showed that the treatment “BSA + trehalose” had the highest arc velocity (1.74μM/s), high
BackgroundSelenium has a major role in male reproduction and antioxidative mechanisms. Although deficiency of this element can result in damages to the body's organs, this metalloid can induce deleterious effects in organisms by causing oxidative stress. This study assessed the spermatotoxicity of selenium nanoparticles (SeNPs) in goldfish (Carassius auratus) based on genotoxicity, antioxidant status, sperm quality, and histopathology.MethodsThe fish with an average weight of 70 g (n = 288) were divided into four experimental groups (three replicates) and fed three times a day with SeNPs at different levels of 0, 0.1, 0.5, and 1 mg kg diet for 30 and 60 days.ResultsAfter 30 and 60 days of feeding trial, compared to the control gro
Sprmatogonial stem cells (SSCs) are valuable for preservation of endangered fish species, biological experimentation, as well as biotechnological applications. However, the rarity of SSCs in the testes has been a great obstacle in their application. Thus, establishment of an efficient in-vitro culture system to support continuous proliferation of SSCs is essential. The present study aimed to establish an efficient and simple method for in vitro culture of Caspian trout undifferentiated spermatogonial cells. Using a two-step enzymatic digestion, testicular cells were isolated from immature testes composed of mainly undifferentiated spermatogonial cells with gonadosomatic indices of <0.05%. The spermatogonial cells were purified by differenti
The present study assessed the protective effect of chitosan-nanoencapsulated quercetin (Qu-ChiNPs) against oxidative stress caused by silver nanoparticles (AgNPs). To this end, the transcription of prime genes regulating hepatic Keap1-Nrf2 pathway as well as downstream antioxidant enzymes were monitored prior to and after oxidative stress by AgNPs. Zebrafish (Danio rerio; n = 225) was assigned into five experimental groups based on feeding with diets supplemented with different additives as follows: negative and positive control groups, without additive; ChiNPs, 400 mg nanochitosan per kg diet; Quercetin, 400 mg free quercetin per kg diet; and Qu-ChiNPs, 400 mg Qu-ChiNPs per kg diet. At the end of the feeding trial (40 days), the
Hepcidins, a group of antimicrobial peptides (AMPs), play a key role in the innate immune system of fishes and act against different pathogens. In this study, antimicrobial and immune-inflammatory activity of a synthetic Ec-hepcidin1, previously identified from orange-spotted grouper, were evaluated. EC-hepcidin1 showed weak activity against the zoonotic fish pathogen Streptococcus iniae (MIC 100 μg mL−1 and MBC 150 μg mL−1). To study the effect of AMPs in general, and Ec-hepcidin1 in particular, a primary cell culture (SC) from the fin tissue of the Caspian Trout (Salmo trutta caspius) was established. The neutral Red method on SC cells revealed that Ec-hepcidin1 has no or very low cytotoxic properties. Treatment of cells wit

Streptococcosis is the most destructive disease in world aquaculture, and many efforts have been made to curb this bacteria. In this study, the ability of artificial polypeptide hepcidin (EC-hepcidin1) was evaluated to control infection caused by Streptococcus iniae in primary cell culture of rainbow trout (Oncorhynchus mykiss). Antibacterial effects of this polypeptide were examined by disk diffusion and minimum inhibitory concentration (MIC) and minimum bactericidal concentration of EC-hepcidin1 were measured against Streptococcus iniae. The primary cells were produced from fish fin, and the toxicity range of the peptide was determined for the cells. The primary cells of the rainbow trout were infected with Streptococcus iniae and antimic
Silver carp (Hypophthalmichthys molitrix) as the major species in total production of freshwater fish, has a special place in the aquaculture industry of Iran. The common method for breeding of this species in our country is semi-natural which provides the brooders opportunities for mate selection that is one of the most striking features in this method. Given the importance of MHC gene complex to sexual behaviors and its historical role in the process of evolution of living organisms especially in terms of safety issues, various patterns of mentioned loci between two generations of Silver carp were studied. 33 males and female silver carp were reproduced in two independent groups by semi-natural method after that larvae in each group were
