Abstract

MIR29B2CHG/C1orf132 is the host gene for generating miR-29b2 and miR-29c. Here, we employed bioinformatics and experimental approaches to decipher expression of C1orf132 in breast cancer cells and tissues. Our data demonstrated a signi cant downregulation of C1orf132 in triple-negative breast cancer. We also predicted a putative promoter for the longer transcripts of C1orf132. The functionality of the distal promoter was con rmed by transfecting MCF7 cells with a C1orf132 promoter-GFP construct. Knocking-out the promoter by means of CRISPR/Cas9 approach revealed no expression alteration of neighboring genes, CD46 and CD34. However, the expression of miR-29c was reduced by half, suggesting an enhancer effect of the distal promoter on miR-29c

Mesenchymal stem cell‐derived extracellular vesicles conditionally ameliorate bone marrow failure symptoms in an immune‐mediated aplastic anemia mouse model

Abstract

Acquired forms of Aplastic anemia (AA) are characterized by T cell‐mediated immune disease resulting in bone marrow (BM) failure and marrow hypoplasia. In these cases, it is a major challenge to modulate autoreactive T cell activity and thereby decrease the pro‐inflammatory cytokine storm. Emerging evidence indicates that extracellular vesicles derived from mesenchymal stem cells (MSC‐EVs) control and modulate immunity. The therapeutic potential of MSC‐EVs has not been investigated in acquired AA. Hence, in this study, we constructed an AA mice model through irradiation and splenocyte infusion to test the benefits of hypoxic MSC‐EVs (Hx‐EVs) and normoxic MSC‐EVs (Nx‐EVs). We found that MSC‐EVs treatment significantly prol

The role of long intergenic non-coding RNA for kinase activation (LINK-A) as an oncogene in non-small cell lung carcinoma

Abstract

The oncogenic role of long intergenic non-coding RNA for kinase activation (LINK-A) has been appraised in triple-negative breast cancer. However, the molecular function of LINK-A is still unclear in most cancers including lung cancer. The present study aimed to evaluate the impact of down-regulation of LINK-A in A549 and Calu-3 cell lines as cellular models of non-small cell lung carcinoma (NSCLC). We used the RNA interference system to knock down LINK-A. LINK-A expression was significantly reduced by siRNA transfection in A549 and Calu-3 cell lines. LINK-A down-regulation significantly reduced cell viability, colony-forming ability and cell migration, as measured by MTT, colony formation and invasion assays. Finally, cell cycle analysis an

Abstract

Familial hypercholesterolemia (FH) is a common, yet underdiagnosed, genetic disorder characterized by lifelong elevated low-density lipoprotein cholesterol levels, which can increase the risk of early-onset coronary artery disease (CAD). In the present study, we screened the nucleotide variations of the LDLR and PCSK9 genes, as well as a part of the APOB gene, in Iranian patients with FH and premature CAD to find the genetic cause of the disorder. Fifteen unrelated individuals with a clinical diagnosis of FH and premature CAD were recruited. Direct DNA sequencing was applied to screen the whole coding exons and exon-intron boundaries of the LDLR and PCSK9 genes and the main parts of their introns, together with exon 26 of the APOB gene. Abstract

Objective: Leishmaniasis is caused by members of the Leishmania species and constitute a group of infective diseases that range from cutaneous lesions to lethal visceral forms. In infected persons, macrophages recognize and eliminate the parasites via phagocytosis. In order to change a hostile environment into an environment adequate for survival and reproduction, the engulfed Leishmania species needs to modulate the function of its host macrophage. The expression patterns of cytokine genes such as interleukin-12 (IL-12), tumour necrosis factor-alpha (TNF-α), IL-1, and interferon-gamma (IFNγ) represent the immune response. In this study, we employed an RNA-seq approach for human monocyte-derived macrophages infected with Leishmania major

Hemophilia A disease: Identification and prediction of new microRNA

Abstract

Various mutations in factor VIII locus are led to an X-linked bleeding disorder in hemophilia A. One of the leading causes of inefficient remedy for hemophilia A disease is the lack of specific and sensitive procedure for punctual diagnosis of the disease. miRNAs indicate that they have a great potential as diagnostic and prognostic biomarkers due to their stability in body fluids and their capability to identification. Furthermore, a relationship between alteration in expression levels of miRNAs and onset and progression of the hemophilia A disease has been reported.Prediction of new miRNAs and nomination as regulators of FVIII gene has been considered as the objective of this study. The ability to express new miRNAs in FVIII locus was stu

Abstract

Obesity is one of the prevalent health-threatening conditions; however, it is preventable by lifestyle interventions such as exercise. The molecular mechanisms underlying physiological adaptation to physical activity are not fully understood. It has been documented that both intracellular and extracellular (circulating) microRNAs (miRNAs) are involved in both obesogenic and exercise adaptation mechanisms. We aimed to conduct a systematic review of publications that examined the effect of exercise on the expression of miRNAs in individuals with obesity. In addition, bioinformatics analysis was performed on most repetitive miRNAs. PubMed, Scopus, and Google Scholar were searched with relevant keywords. We only included studies that utilized e

Abstract

In this report, we look at the challenges posed by the outbreak of COVID‐19 and how the Executive Board of these two congresses succeeded in overcoming those challenges and holding two congresses. The approach for a large festival with different virtual setting components provided a suitable solution that led to exemplary achievements and created an appropriate model for future virtual or combined virtual and face‐to‐face events. These events proved that pandemic problems could not limit the organizers, pushing them to make better use of the facilities and turning this threat into an opportunity.
